Due to the high demand for village chickens in Malaysia, there is a concern that commercial chickens, due to their cheap cost, are claimed as the village chicken to fraud consumers for economic gain since there is no strategy to authenticate different chicken breeds in Malaysia. According to several studies, the most reliable method for differentiating among different breeds of animals is a genomic study to understand genome differences, and therefore the discovery of genomic biomarkers to facilitate discrimination of different chicken breeds is of paramount importance.
Therefore, in the present invention, the simple and low-cost novel nucleic acid-based lateral flow assay (NALFA) is developed using a breed-specific genomic region with high discriminating power for rapid and sensitive authentication of Malaysian village chicken through combining of isothermal rapid isothermal recombinant polymerase amplification (RPA) technique.
The unique aspect of this work will be its novelty regarding the genomic region that can consider as a biomarker to differentiate village chicken from non-village chicken breeds. Moreover, this developed NALFA will be the first assay for the purpose of chicken breed authentication which can be beneficial for the industry for on-site detection of food frauds in Malaysia.
Another advantage of the present invention is applying the RPA which is a single tube, an isothermal alternative to the conventional polymerase chain reaction (PCR). RPA is low-cost, time-saving, and operationally simple that just takes just 20 minutes to complete and doesn’t need a thermocycler like conventional PCR.
Invention
The present invention relates to a nucleic acid-based lateral flow assay (NALFA) strip for the detection of village chickens from non-village chicken breeds in Malaysia.
The unique aspect of this work will be its novelty regarding the breed-specific genomic region that can consider as a biomarker to differentiate village chicken from non-village chicken breeds.
Compared to conventional lateral flow immunoassays which are based on expensive antibodies, the proposed method by applying a novel breed-specific genomic region to fabricate nucleic acid lateral flow assay can be used to rapidly distinguish village chickens over non-village chicken breeds, which is not only accurate in identification; it has simple operation, sensitive and fast detection, and can save cost.
Another advantage of the present invention is applying the Recombinase polymerase amplification (RPA) which is a single tube, an isothermal alternative to the conventional polymerase chain reaction (PCR) and therfore can save assay time and cost. It should be noted that the lateral flow strips used in this study are recycable.
